Introduction:
We have developed an integrated antibody humanization and affinity maturation platform based on our advanced Phage/ Yeast display technology and our computer modeling and simulation techniques. Through combining the methods of bioinformatics and structural biology, including in silico CDR-grafting, computer-aided design and optimization, high-throughput screening of Phage-display antibodies, Biacore surface resonance analysis and so on, we are able to provide humanized antibodies with diminished immunogenicity but comparable or better specificity, bioactivity, thermos-stability and productivity than the parental antibodies. The binding affinity of the humanized antibodies can also be further improved at least 5 folds by utilizing our affinity maturation technology.
